RESUMO Este estudo avaliou os efeitos da exposição ao manganês (Mn2+), após 96 horas, a duas temperaturas (24 e 27°C) em juvenis de Centropomus parallelus por meio de análises bioquímicas (atividade das enzimas glutationa S-transferase (GST) e catalase (CAT)) e genotóxicas (teste do micronúcleo e ensaio cometa). A atividade da GST não mostrou diferença significativa entre os grupos expostos ao Mn2+ e os seus respectivos grupos controle, enquanto que um aumento significativo na atividade da CAT foi observado a 27°C no grupo exposto ao Mn2+, quando comparado ao grupo controle. As análises genotóxicas mostraram que os animais expostos ao Mn2+ tiveram aumento significativo na quantidade de células com micronúcleo em relação aos seus grupos de controles. Houve também aumento significativo na incidência de danos ao DNA nos grupos expostos a esse contaminante. Na temperatura de 24°C, os animais expostos ao Mn2+ tiveram maior quantidade de danos no DNA em relação a 27°C. É provável que o aumento da temperatura também possa induzir o estresse oxidativo. Assim, concluímos que o manganês é tóxico para os juvenis de robalo, causando dano genotóxico, e quando associado a um aumento da temperatura, também pode provocar um aumento no estresse oxidativo.

ABSTRACT This study evaluates the effects of exposure to manganese (Mn2+) for 96 hours at two different temperatures (24 and 27°C) on juveniles of Centropomus parallelus through the activities of glutathione S-transferase (GST) and catalase (CAT), micronuclei test (MN) and comet assay. The GST activity did not show any significant difference between the groups exposed to Mn2+ and the respective control groups; in contrast, a major increase in the CAT activity was observed at 27°C in the group exposed to Mn2+ compared to the control group. The genotoxic analyses showed that in all animals exposed to Mn2+, the number of red cells with micronuclei increased significantly compared to the respective control groups. There was also a significant increase in the incidence of DNA damage in the groups exposed to Mn2+. At a temperature of 24ºC, animals exposed to Mn2+ had more DNA damage than those at 27°C. It is likely that the increase in temperature can also induce oxidative stress. Thus, we conclude that manganese is toxic to the fat snook juveniles, causing genotoxic damage, and when associated with an increase in temperature, manganese can also provoke an increase in oxidative stress.

O objetivo deste trabalho foi avaliar o efeito da temperatura e da taxa de alimentação sobre o crescimento e a produtividade de juvenis de robalo-peva (Centropomus parallelus). Utilizou-se experimento fatorial com duas temperaturas (25 e 28º C) e duas taxas de alimentação (3 e 6% da biomassa ao dia), com três repetições para cada combinação de temperatura e taxa. Os peixes (9,80±0,41 g; 9,86±0,14 cm) foram alimentados duas vezes ao dia durante 60 dias. A cada 15 dias, foram realizadas amostragens para corrigir a quantidade de ração oferecida. Não houve mortalidade durante o experimento. Maior crescimento em peso e comprimento foi obtido nos animais submetidos à temperatura de 28ºC. Ganho de peso, biomassa final, conversão alimentar aparente e taxa de crescimento específico foram influenciados pela taxa de alimentação e pela temperatura. A oferta de 3% da biomassa ao dia a 28ºC proporciona melhores índices de crescimento e produtividade para juvenis de robalo-peva.

The objective of this work was to evaluate the effects of temperature and feeding rates on growth and productivity of juvenile fat snook (Centropomus parallelus). A factorial experiment with two temperatures (25 and 28ºC) and two feeding rates (3 and 6% of biomass per day) was used, with three replicates for each combination of temperature and rate. Fish (9.80±0.41 g; 9.86±0.14 cm) were fed twice a day during 60 days. Every 15 days, samples were taken to correct the amount of food offered. No mortality was observed during the experiment. A greater growth in weight and length was obtained in animals subjected to the temperature of 28ºC. Weight gain, final biomass, apparent food conversion, and specific growth rate were influenced by feeding rate and temperature. The offer of 3% of biomass per day at 28ºC provides better growth and productivity indices for juvenile fat snook.

O óleo diesel pode ser uma fonte de contaminação em ambientes aquáticos, principalmente como resultado de derrames acidentais. Foram avaliados os efeitos da fração solúvel do óleo diesel (FSO) em Prochilodus vimboides. Os peixes foram expostos a três diferentes diluições da FSO por até 96 horas e comparados com um grupo controle. Os danos nos fragmentos de DNA foram analisados utilizando o ensaio Cometa. A presença de anormalidades nos eritrócitos foi avaliada pelo teste do micronúcleo. A atividade da Aspartato aminotransferase (AST) e alanina aminotransferase (ALT) e da acumulação de cobre nas brânquias também foram analisados. Os peixes expostos por 96 h tiveram maiores taxas de danos do que aqueles expostos por 24 h. Não houve diferença significativa quanto à presença de micronúcleos entre os peixes expostos e controle e entre 24 e 96 h exposições. Para AST, não foi observada diferença significativa entre as amostras coletadas em dois tempos de exposição. Os peixes expostos a uma diluição de 1:100 do FSO apresentaram maior atividade da enzima ALT do que os peixes do controle após um período de exposição de 24 horas. Não houve bioacumulação de cobre nas brânquias. Os efeitos genotóxicos nas células foram os mais evidentes em P. vimboides durante a exposição aguda a FSO.

Diesel oil can be a source of contamination in aquatic environments, mainly as a result of spills. The effects of the water-soluble fraction of diesel (WSF) on Prochilodus vimboideswere assessed. Fish were exposed to three different WSF dilutions for up to 96 h and were compared to a control group. Damages in the fragments of DNA were analyzed using the Comet assay. The presence of erytrocyts abnormalities was assessed by micronucleus test. Aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity and the accumulation of copper in gills were also analyzed. Fish exposed for 96 h had higher rates of damage than those exposed for 24 h. There was no significant difference regarding the presence of micronuclei between exposed and control fish and between 24-h and 96-h exposures. For AST, no significant difference was observed between samples collected at the two exposure times. Fish exposed to a 1:100 dilution of WSF showed higher activity of the enzyme ALT than the control fish after a 24-h exposure period. There was no bioaccumulation of copper in the gills. We conclude that the genotoxic effects of WSF in the cells are more evident in P. vimboides during an acute exposure.

The present study evaluated the sensory, proximate and morphometric differences of pond and net-cage-reared Nile tilapia (Oreochromis niloticus). Thirty samples from ponds and 30 samples from net-cages were used. Morphometric measurements were performed in fish and the fillets; proximate composition of the fillets was determined; and sensory analysis of the meat was performed. The fish reared in ponds exhibited similar weights and larger measurements than those reared in net-cages. Fillet measurement and weight were similar for both rearing techniques. The proximate composition analysis of the fillets showed that there was less lipid deposition (12.6 g/kg) and higher moisture retention (792.6 g/kg) in fish reared in the ponds compared with those reared in net-cages (31.7 and 767.8 g/kg, respectively). Although the amount of lipid in the fillets from fish reared in net-cages was higher, the fillets do not contain excessive fat. There was no difference in fillet protein or ash. The flavor of the fillet was moderately good to good and was similar in both farming systems. Fillets produced with the 2 farming systems have similar morphometric, proximate and sensory characteristics.

A inevitável manipulação de peixes vivos em piscicultura causa uma série de reações fisiológicas adversas, o que exige o uso de anestésicos. O objetivo foi avaliar a eficiência do óleo de cravo como anestésico para juvenis avançados (aproximadamente 55 g e 13 cm) de tilápia do Nilo durante o manejo. No primeiro experimento, os peixes foram expostos a banhos anestésicos em seis diferentes concentrações (80; 100; 150; 200; 250 e 300 mg L-1) e foi avaliado o tempo de indução aos diferentes estágios de anestesia. No segundo experimento, avaliaram-se os diferentes tempos de exposição à anestesia, sendo verificada a margem de segurança do anestésico. Por último, avaliaram-se as respostas de estresse após a anestesia em 250 mg L-1. Os parâmetros avaliados foram cortisol, glicose, hematócrito, hemoglobina e níveis plasmáticos de sódio e potássio. A concentração 250 mg L-1 de óleo de cravo foi a mais adequada para indução de anestesia cirúrgica. Para a anestesia voltada para biometria e breve manejo, a concentração mais adequada foi 150 mg L-1. A exposição à concentração ideal de óleo de cravo por 10 min., não causou estresse severo, ocorrendo alterações após a anestesia somente nos níveis de glicose e hematócrito.

Handling of live fish is inevitable in fish farms and causes a number of adverse physiological reactions, thus requiring the use of anesthetics. The main goal of the present work was to evaluate the efficiency of clove oil as an anesthetic for juvenile tilapia (approximately 55 g and 13 cm) during handling. In the first experiment, fishes were exposed to an anesthetic bath at six concentrations (80, 100, 150, 200, 250 and 300 mg L-1). Subsequently, the induction time was assessed for different stages of anesthesia. In the second experiment, different times of exposure to anesthesia were tested, aiming to assess the safety margin of the anesthetic. Finally, we evaluated the responses to stress after exposure to 250 mg L-1 anesthesia. The following parameters were tested: cortisol, glucose, hematocrit, hemoglobin, and plasma sodium and potassium levels. The concentration of 250 mg L-1 of clove oil is the most appropriate to induce surgical anesthesia, and 150 mg L-1 for biometrics and brief handling. Anesthesia at the ideal concentration for 10 minutes did not result in severe stress. Only blood glucose and hematocrit were affected after anesthesia.

The high toxicity of diesel components makes oil spills a threat to the biota in coastal marine environments. The genotoxic effect of the diesel water-soluble fraction (DWSF) on Hippocampus reidi (Ginsburg, 1933) was assessed. Fish were exposed to three different DWSF dilutions for up to 96 hours, and genotoxicity was analyzed using the micronuclei test and the comet assay. The micronuclei test revealed no significant differences between any of the DWSF dilutions and the control in the 24-hours period; however, micronuclei increased in fish exposed to 1:500 and 1:100 DWSF dilutions for 96 hours. For all dilutions, there was an increase in micronuclei in fish exposed for 96 hours when compared to those exposed for 24 hours. The tested dilutions increased frequencies of cell classes 2 (medium damage) and 3 (large damage) in the comet assay after 24 and 96 hours. Fish exposed to DWSF 1:100 exhibited a higher frequency of class 4 (apoptosis) cells in the 96-hours period. All dilutions increased the comet score when compared to the control at 24 and 96 hours. The micronuclei and comet tests were efficient in detecting DWSF genotoxic effects in H. reidi.

Os peixes que vivem na Amazônia são expostos a vários tipos de água: águas pretas, contendo grande quantidade de carbono orgânico dissolvido, águas brancas, com concentração de Ca2+ dez vezes maior que as águas pretas e pH neutro, e águas claras, com concentração de Ca2+ duas vezes maior que as águas pretas e pH também neutro. Dessa forma, o objetivo deste trabalho foi analisar o fluxo de Ca2+ no peixe de respiração aérea facultativa Hoplosternum littorale (tamoatá) exposto a diferentes tipos de águas amazônicas. Os peixes foram aclimatados em água de poço artesiano (semelhante à água clara) e depois colocados individualmente em câmaras para medir o fluxo de Ca2+. Após 4 h, a água das câmaras foi trocada por um tipo diferente de água. A transferência do tamoatá das águas pobres em íons água preta e preta ácida ou da água branca, rica em íons, para as águas preta e preta ácida, pobres em íons, resulta em uma perda de Ca2+ apenas nas duas primeiras horas de experimento. Entretanto, a transferência da água preta e preta ácida, para a água branca resulta em um influxo de Ca2+. Os resultados obtidos nos permitem concluir que a transferência do tamoatá para as águas preta e preta ácida, pobres em íons, leva a uma temporária perda de Ca2+, e a quantidade de Ca2+ na água branca, rica em íons, é adequada para prevenir sua perda após a transferência. Sendo assim, a transferência do tamoatá entre as águas estudadas não resulta em sérios distúrbios no Ca2+.

Fishes that live in the Amazonian environment may be exposed to several kinds of waters: "black waters", containing high dissolved organic carbon and acidic pH, "white waters", with ten fold higher Ca2+ concentrations than black waters and neutral pH, and "clear waters", with two fold higher Ca2+ concentrations than black waters and also neutral pH. Therefore, the aim of the present study was to analyze Ca2+ fluxes in the facultative air-breather Hoplosternum littorale (tamoatá) exposed to different Amazonian waters. Fishes were acclimated in well water (similar to clear water) and later placed in individual chambers for Ca2+ fluxes measurements. After 4 h, water from the chambers was replaced by a different type of water. Transfer of tamoatás to ion-poor black or acidic black water resulted in net Ca2+ loss only in the first 2 h of experiment. However, transfer from black or acidic black water to white water led to only net Ca2+ influxes. The results obtained allowed us to conclude that transfer of tamoatás to ion-poor waters (black and acidic black water) led to transient net Ca2+ loss, while the amount of Ca2+ in the ion-rich white water seems adequate to prevent Ca2+ loss after transfer. Therefore, transfer of tamoatás between these Amazonian waters does not seem to result in serious Ca2+ disturbance.

Foram determinadas as temperaturas letais (TL50) inferiores e superiores de larvas de R. quelen. As larvas foram obtidas em dois períodos de desova com temperatura de incubação de 21 e 26ºC, respectivamente. Larvas foram colocadas em aquários onde a temperatura foi elevada em 1ºC, a cada 15min, ou reduzida 1º C, a cada 20min, até as larvas cessarem seus movimentos. Esta temperatura foi então fixada e mantida por 96h. O mínimo de mortalidade foi de 29 ± 1,5% obtido nas temperaturas de incubação. As TL50 inferiores foram 15,01 e 16,72ºC e as TL50 superiores foram 29,25 e 27,83ºC para as larvas incubadas a 21 e 26ºC, respectivamente. Portanto, aparentemente, não há diferença com relação à tolerância a variações de temperatura nas larvas incubadas nas duas temperaturas.

The lower and upper lethal temperatures (LT50) of R. quelen larvae were determined. The larvae were obtained from two spawning periods with hatching temperatures of 21 and 26ºC respectively. Larvae were placed in tanks at which the temperatures were either increased 1ºC every 15min, or reduced 1ºC every 20min until larval movement ceased. This temperature was then maintained for 96h. Minimum mortality (29.0 ± 1.5%) was observed in the larvae held at hatching temperatures. The lower LT50 were 15.01 and 16.72ºC, and the upper LT50 were 29.25 and 27.83ºC for the larvae hatching at 21 and 26ºC, respectively. Thus, apparently there is no difference with relation to temperature change tolerance between larvae hatched in both temperatures.
